Addex Pharmaceuticals / Addex: ADX71943 Demonstrates Analgesic Effects in
Pain Models processed

Addex Pharmaceuticals (SWISS: ADXN), the
allosteric modulation company, announced today that ADX71943 has
demonstrated
statistically significant analgesic-like effects in three preclinical
models of
pain.
Inthe firstmodel, ADX71943reversed completeFreund adjuvant (CFA)-
induced
reductionsin withdrawalthresholds ina dose-relatedmanner, with a
minimum
statisticallysignificant effectiveoral doseof 10 mg/kg.In a second
model
(Formalin test), ADX71943 showed anti-nociceptive effect after
oral
administrationof 3 to 10 mg/kg.In a thirdmodel, an increased visceral
pain
thresholdwasseenusingADX71943witha minimum statistically
significant
effective dose between 3 and 10 mg/kg.
ADX71943isanorallyavailablepositiveallosteric modulator (PAM)
of the
GABA(B)receptor thathas potentialfor treatmentof osteoarthritis
pain and
chronicnociceptivepainaswellasother indications. ADX71943 has a
good
developabilityprofile and is expected toenter clinical testing in the
fourth
quarter 2010.
"We are impressed with the profile of the compound and by its activity in
these
models. Furthermore,
minimum effective doses observed in thepreclinicalmodelsand
the
pharmacokineticpropertiesofthecompoundsupportprediction of
treatment
regimensin humans compatiblewith the targetpatient population and
disease"
said Sonia Poli, head of non-clinical development at Addex.
GABA(B)activationisaclinicallyvalidated mechanism. Baclofen, a
GABA(B)
agonist,is widely usedfor its musclerelaxant properties inthe
control of
spasticitycaused by multiple sclerosis, cerebralpalsy or certain
injuries to
thespine. Furthermore, Baclofen is a powerful analgesic agent, especially
when
delivered intrathecally. However, rapid induction of tolerance and dose
limiting
CNS-mediatedsideeffects(e.g.sedation,hypothermia,memory
impairment)
preventbaclofenfrombeingwidelyprescribedforpain relief
despite its
analgesicproperties. ADX71943,an allostericmodulator with a
differentiated
profile,couldavoidthetoleranceanddose limiting side effects
commonly
associated with baclofen.
Osteoarthritis(OA) is a widespreadcondition affecting the elderly
population
inparticular. One of the most debilitating symptoms is chronic pain
associated
withOA. Patients sufferingfrom chronic painwill require treatment
over the
long-term. There is a clear need for drugs with better side effect profiles
than
current marketed drugs (e.g. non-steroidal anti-inflammatory drugs and
opioids).
