Addex Pharmaceuticals (SWISS: ADXN), the allosteric modulation company, announced today that ADX71943 has demonstrated statistically significant analgesic-like effects in three preclinical models of pain.

Inthe firstmodel, ADX71943reversed completeFreund adjuvant (CFA)- induced reductionsin withdrawalthresholds ina dose-relatedmanner, with a minimum statisticallysignificant effectiveoral doseof 10 mg/kg.In a second model (Formalin test), ADX71943 showed anti-nociceptive effect after oral administrationof 3 to 10 mg/kg.In a thirdmodel, an increased visceral pain thresholdwasseenusingADX71943witha minimum statistically significant effective dose between 3 and 10 mg/kg.

ADX71943isanorallyavailablepositiveallosteric modulator (PAM) of the GABA(B)receptor thathas potentialfor treatmentof osteoarthritis pain and chronicnociceptivepainaswellasother indications. ADX71943 has a good developabilityprofile and is expected toenter clinical testing in the fourth quarter 2010.
